چکیده

Title of Document: EVALUATION OF LEF1 TRANSCRIPTION FACTOR IN MAMMALIAN PREIMPLANTAITON EMBRYOS Ashley Elaine Meece, Master of Science, 2007 Directed By: Associate Professor, Dr. Carol L. Keefer Department of Animal and Avian Sciences The first lineage differentiation of cells during pre-implantation embryo development is critical for continued embryonic growth. Though several transcription factors (TFs) have been identified that are involved in this transition, a specific mechanism of regulation has yet to be determined. Previous studies in our laboratory have shown the TF Lef1 is involved in differentiation of mouse embryonic stem cells into trophoblastic stem cells through the Wnt signaling pathway. Lef1 is known to be involved in lineage determination of adult skin stem cells. As we observed that two isoforms of Lef1 were expressed at different stages of pre-implantation development, we hypothesized that Lef1 was interacting with the other well-established lineage differentiation TFs, Nanog, Oct4, and Cdx2, in these early embryos. At the blastocyst stage, no significant changes in mRNA expression were seen when siRNAs, specifically designed to knockdown Lef1 expression, were injected in early embryos; however, knockdown of Lef1 did not interfere with blastocyst formation. Hippocampus development and generation of dentate gyrus granule cells is regulated by LEF1.

Lef1 and other genes of the LEF1/TCF family of transcription factors are nuclear mediators of Wnt signaling. Here we examine the expression pattern and functional importance of Lef1 in the developing forebrain of the mouse. Lef1 is expressed in the developing hippocampus, and LEF1-deficient embryos lack dentate gyrus granule cells but contain glial cells and interneurons in the region of the de...

Lymphoid enhancer binding factor 1 regulates transcription through gene looping.

Efficient transcription depends upon efficient physical and functional interactions between transcriptosome complexes and DNA. We have previously shown that IL-1beta-induced lymphoid enhancer binding factor 1 (Lef1) regulates the transcription of its target genes COX2 and MMP13 in mouse chondrocytes by binding to the Lef1 binding sites located in the 3' region. In this study, we investigated ho...

I-13: Transcriptome Dynamics of Human and Mouse Preimplantation Embryos Revealed by Single Cell RNA-Sequencing

Background: Mammalian preimplantation development is a complex process involving dramatic changes in the transcriptional architecture. However, it is still unclear about the crucial transcriptional network and key hub genes that regulate the proceeding of preimplantation embryos. Materials and Methods: Through single-cell RNAsequencing (RNA-seq) of both human and mouse preimplantation embryos, ...

A yeast model system for functional analysis of β-catenin signaling

We have developed a novel Saccharomyces cerevisiae model system to dissect the molecular events of beta-catenin (beta-cat) signaling. Coexpression of mammalian beta-cat with TCF4 or LEF1 results in nuclear accumulation of these proteins and a functional complex that activates reporter gene transcription from constructs containing leukocyte enhancer factor (LEF)/T cell factor (TCF) response elem...

Evaluation of the Effects of Nicotine on Mammalian Target of Rapamycin Complex 2 and Signal Transducer and Activator of Transcription 3 Genes Expression in a Mouse Model of Allergic Asthma: An experimental study

Background & Aims: Allergic diseases have increased in the last decade worldwide and researchers have been trying to introduce new strategies and drugs to treat these types of diseases. Nicotine shows anti-inflammatory properties and the studies have revealed that it can reduce the inflammation and the allergic responses. The mammalian target of rapamycin (mTOR) is a multifunctional protein kin...
